24 hour urine collection method, Urinary total protein examination

24 hours urine collection method:

  • Take a clean container. (In which it is possible to take urine for 24 hours).
  • Before taking urine in bottle 10-15 ml toluene or 1.5 ml formaldehyde or a piece of thymol or 50 drops chloroform or 50 gm boric acid should be given 
  • Then you have to collect urine in this container for 24 hours.
  • In case of 24 hours urine collection, the amount of urine for 24 hours should be determined with the help of measuring cylinder and other necessary tests should be done.

24 hours total urinary protein examination:

  • After Collecting 24 hours urine then determine the amount.
  • If the urine is alkaline, mix a few drops of 5% acetic acid with urine
  • The reaction of urine should be made acidic.
  • Urine should be taken up to the U-spot of Esbachs albuminometer.
  • After taking the urine, the albuminometer should take Esbachs reagent till the stain.
  • Close the mouth of the albuminometer well and shake the albuminometer tube up and down slowly to mix urine and reagent well.
  • The AIbuminometer tube should be kept upright in a safe place for 24 hours.
  • After 24 hours, the bottom of the albuminometer will need to determine how many spots of white protein sediment have accumulated.
  • The amount of protein in the urine per liter (gm / l)
  • If sediment accumulates up to 1 spot of AIbuminometer tube, then 1g / L should be written in the report or patients should determine the amount of protein in 24 hours by calculating the amount of urine in 24 hours.

Calculation:

24 Hours urinary protein = g / L protein / 1000x24 hours urine (m)


Esbach's Reagent made
Picric acid ........................................ 10 gram
Citric acid ........................................ 20 grams
Distilled water .................................. 900 ml
Boiling and mixing. The total volume should be 1000m1 by mixing distilled water.
